The city's property and development committee voted 2-1 on Monday to ignore a recommendation that would have protected the three-storey building.
The owner wants to demolish the Annex to make way for a 275-stall parkade that would also cover an adjacent empty lot. The plan will not affect the 103-year-old Grain Exchange Building, which is connected to the Annex via a second-floor bridge.
Couns. Scott Fielding (St. James) and Mike O'Shaughnessy (Old Kildonan) rejected the status, while Russ Wyatt (Transcona) wanted to see it preserved.
For demolition to proceed, the decision must be affirmed by executive policy committee on Wednesday and council on July 22.
